
High-Throughput Purification of Peptides and PNAs Simplified

Chemical synthesis of fluorescently labelled and not
labeled peptides or peptide nucleic acids (PNAs) has been scaled down to the multi-well plate format and below, making the generation of large peptide libraries possible. After synthesis, removal of side products, e.g. premature termination products, is mandatory in order to obtain highly purified products. This purification is typically performed using HPLC, a method which is not suited for high-throughput purification. So, as yet, purification is the bottleneck for the generation of complex peptide or PNA libraries consisting of purified components. Here, we offer a technology to purify full-length products from synthesis mixtures without the need for HPLC, opening the way for a low-cost generation of highly purified and complex peptide and PNA libraries for highthroughput screening.
