Festphasensynthese Trinukleotide
Alle bisherigen bekannten Verfahren zur Synthese von Trinukleotidsynthons verlaufen in der
Lösungschemie nach dem Phosphortriesterverfahren oder dem Phosphoramiditverfahren. Diese mehrstufigen Verfahren besitzen jedoch den Nachteil, dass sie sehr zeitaufwendig sind, da nach jedem Syntheseschritt eine Isolation und Reinigung des Produktes notwendig ist. Durch die Festphasenchemie lassen sich diese sehr zeitaufwendigen Schritte umgehen. Bei der Festphasenchemie ist lediglich ein Herun-terwaschen der Nebenprodukte, Reagenzien und unumgesetzten Ausgangsstoffe vom Träger erforderlich, während das gewünschte Produkt auf der Festphase verbleibt. Dies erlaubt eine schnelle Synthese der Trinukleotide schon in wenigen Stunden.
