Antisense tools for functional studies of human Argonaute proteins

Specific antisense oligonucleotides were developed for the four human Argonaute proteins as non-RNAi based tools for the knockdown of human Ago expression on the mRNA and protein levels. As antisense oligonucleotides make use of RNase H based degradation of target mRNA, which is distinct from the RNAi pathway and does not interfere with RNAi or its constituents, this alternative and more unbiased antisense oligonucleotide-based approach is a choice alternative to study the biology of the different human Argonaute species as well as for the therapy of diseases based on biological processes involving Argonaute proteins on the molecular level.
